DIPK2B (divergent protein kinase domain 2B) is an X-linked gene encoding a predicted calcium-regulated protein kinase 1. The gene belongs to the FAM69 family of kinases, which contain conserved catalytic motifs suggesting retained phosphotransferase activity and an EF-hand calcium-binding domain indicative of calcium-dependent regulation 1. DIPK2B likely functions in phosphorylation of proteins within the secretory pathway and/or extracellular proteins, given the presence of signal peptides in FAM69 family members 1. At the molecular level, DIPK2B expression is associated with incubation of methamphetamine craving in animal models, with increased striatal Dipk2b mRNA levels correlating with compulsive drug-seeking behavior in shock-resistant rats 2. Genetically, DIPK2B variants are associated with the Castelli risk index 2 (LDL-C/HDL-C ratio), a predictor of cardiovascular risk 3, suggesting a role in lipid metabolism regulation. Clinically, DIPK2B is linked to X-linked mental retardation (XLMR) and autism spectrum disorder (ASD)-like traits through its relationship to the DIA1R gene family, which shows point mutations and deletions associated with these neurodevelopmental conditions 4. The gene's neurological involvement and lipid metabolism associations suggest its dysfunction may contribute to both neuropsychiatric and metabolic disorders.
