ERVH48-1 (Suppressyn/SUPYN) is a primate-specific endogenous retroviral envelope protein with dual roles in development and host defense. Primarily, ERVH48-1 functions as a negative regulator of trophoblast syncytialization by binding to SLC1A5 (the receptor for syncytin-1/ERVW-1) and inhibiting cell-cell fusion 1, a mechanism critical for controlling placental morphogenesis 2. Beyond placentation, ERVH48-1 regulates somatic cardiomyocyte development by binding the WNT antagonist SFRP2, promoting its polyubiquitination and degradation to enable WNT/β-catenin signaling 3. Additionally, ERVH48-1 enhances somatic cell reprogramming efficiency, facilitating iPSC generation with full germ layer differentiation capacity 4. At the immunological level, ERVH48-1 demonstrates antiviral activity, restricting infection by mammalian type D retroviruses—evidence of host genome domestication for viral defense 5. Clinically, ERVH48-1 expression is altered in breast cancer and may function as a tumor suppressor 6. ERVH48-1 is expressed in human preimplantation embryos, developing placenta, and trophoblast cells, with localization to subcellular membrane compartments via an N-terminal signal peptide 37. Dysregulation of ERVH48-1 may contribute to adverse pregnancy outcomes and disease pathology.
