FAM168B (family with sequence similarity 168 member B), also known as myelin-associated neurite-outgrowth inhibitor (MANI), is a membrane-associated protein that functions as a negative regulator of neuronal axonal outgrowth 1. The protein inhibits axonal growth through interaction with cell division cycle protein 27 (CDC27), a component of the anaphase-promoting complex (APC), establishing the MANI-CDC27-APC pathway as a critical cascade preventing neuronal axon extension 1. FAM168B also acts as a negative regulator of CDC42 and STAT3 signaling while positively regulating STMN2 and CDC27. Clinically, FAM168B deletions are associated with neurodevelopmental disorders; rare recurrent microdeletions at chromosome 2.1 encompassing FAM168B correlate with developmental delay, intellectual disability, attention-deficit hyperactivity disorder (ADHD), epilepsy, and aggressive behavior 23. Beyond neurology, FAM168B has emerged as a novel candidate target for chimeric antigen receptor (CAR) T cell-based cancer immunotherapy, with expression on cancer cell surfaces suggesting potential as a tumor-specific therapeutic antigen 4. Additionally, gene co-expression network analysis identified FAM168B as a hub gene associated with Salmonella infection resistance in poultry, implicating its role in immune response pathways 5.
