Galanin (GAL) is a neuropeptide hormone of the central and peripheral nervous systems that binds and activates the G protein-coupled receptors GALR1, GALR2, and GALR3. The peptide is widely distributed in neural tissues, including motor neurons where it appears early in development alongside other neuropeptides 1, and in sensory and autonomic nerve fibers throughout the gastrointestinal and genitourinary tracts 23. Galanin exhibits molecular heterogeneity across species, with multiple forms detected chr11 in humans 23. Functionally, galanin modulates smooth muscle contraction and neurotransmission; in the human urinary bladder, it potently inhibits acetylcholine-induced contractions in a concentration-dependent manner 4, suggesting a neuromodulatory role in regulating genitourinary tract motility and tone. Galanin-immunoreactive nerve fibers are localized around eccrine sweat glands and in close association with blood vessels in the genitourinary system 53, consistent with broader physiological roles in smooth muscle regulation, blood flow, and hormone secretion. Clinical relevance of GAL dysfunction is suggested by its association with epilepsy, skeletal abnormalities, and various malignancies, though mechanistic details remain to be elucidated.
