KNDC1 is a brain-specific RAS guanine nucleotide exchange factor (GEF) that regulates multiple cellular processes with emerging clinical relevance. Structurally, KNDC1 functions as a putative protein-protein interaction module localized to dendrites and guanine nucleotide exchange factor complexes in neuronal cell bodies 1. Its primary neuronal function involves negative regulation of dendrite growth and morphogenesis 2. Beyond neurobiology, KNDC1 plays a critical role in cellular senescence. KNDC1 knockdown delays endothelial cell senescence by promoting proliferation through the ERK signaling pathway while inhibiting the p53-p21-p16 cascade 2. Conversely, KNDC1 overexpression promotes senescence via a p53-ROS positive feedback loop 1. In ovarian cancer, KNDC1 functions as a tumor suppressor, with downregulation associated with enhanced ERK1/2 phosphorylation and increased proliferation; lower KNDC1 expression predicts poor progression-free survival 3.
