LHFPL2 is a tetraspan membrane protein with diverse roles in development and disease. While it is known to play a role in female and male fertility and distal reproductive tract development [UniProt annotation], recent genetic studies have revealed unexpected associations with neurodegenerative and malignant diseases. A genome-wide association study identified LHFPL2 variants on chromosome 5.1 as significant modifiers of age-at-onset in familial Parkinson's disease, with carriers experiencing disease onset approximately 9.79 years earlier than non-carriers 1. In cancer biology, LHFPL2 emerges as a prognostic and immunological factor. In hepatocellular carcinoma, LHFPL2 is part of an mTORC1 pathway-related signature associated with poor prognosis, immune evasion, and reduced immunotherapy response 2. In renal cell carcinoma, LHFPL2 upregulation correlates with poor survival and high tumor stage, and positively correlates with M2 macrophage polarization and immune checkpoint expression, suggesting a role in immunosuppression 3. Additionally, LHFPL2 expression increases with cellular senescence and age in peripheral blood 4, and it has been associated with subclinical atherosclerosis and coronary heart disease in African Americans 5. LHFPL2 is also overexpressed in acute myeloid leukemia with long-term proliferation 6. The specific molecular mechanisms underlying these diverse associations remain to be elucidated.
