Based on limited published evidence, MARCHF4 is an E3 ubiquitin-protein ligase localized to the Golgi stack and trans-Golgi network that mediates ubiquitination of MHC-I and CD4, promoting their endocytosis and lysosomal sorting via multivesicular bodies. MARCHF4 possesses ubiquitin-protein transferase activity and protein binding capacity. In lung adenocarcinoma, MARCHF4 was identified as a key prognostic gene associated with glutathione metabolic reprogramming and immune suppression, with upregulation observed in LUAD cells 1. The gene's role in immune regulation and potential therapeutic response prediction warrants further investigation.
