Based on limited published evidence, NUDT22 is a Mg2+-dependent NUDIX hydrolase that cleaves UDP-glucose and UDP-galactose to produce glucose 1-phosphate (or galactose 1-phosphate) and UMP 1. The enzyme features a unique structural fold with a modified NUDIX domain and N-terminal UDP-binding region. Functionally, NUDT22 promotes pyrimidine salvage in cancer cells by maintaining UMP supply, supporting DNA replication and preventing replication stress 2. NUDT22 expression is elevated in cancer tissues and p53-regulated upon metabolic stress or DNA damage, with high expression correlating with poor survival outcomes.
