RAB33A is a small GTPase belonging to the RAS oncogene family that regulates intracellular membrane trafficking and vesicular transport 1. The protein is primarily expressed in brain tissue, lymphocytes, and normal melanocytes, where it colocalizes with melanosomal proteins and regulates their transport to melanosomes 1. RAB33A functions as a T cell regulatory molecule, with expression predominantly in CD8+ T cells that is induced upon T cell activation 2. In cervical cancer, RAB33A promotes metastasis through a non-canonical autophagy mechanism, where it stabilizes RhoC protein by impairing autophagosome-lysosome fusion via inactivation of RAB7 through interaction with TBC1D2A 3. The gene serves as a potential biomarker for tuberculosis, as RAB33A expression is significantly downregulated in patients with TB disease compared to both latent TB infection and healthy controls 42. RAB33A expression is also suppressed in melanoma through DNA methylation of its promoter region, representing an early event in melanocytic transformation 1.
