SUSD5 (sushi domain containing 5) is a cell anchorage-related protein implicated in multiple cancer-related processes. Based on its GO annotation, SUSD5 participates in Notch signaling pathway regulation 1. In functional studies across cancer types, SUSD5 knockdown impaired gastric cancer cell migration, invasion, and proliferation, with reduced tumor growth and metastasis in vivo, possibly through cAMP signaling modulation 2. In colon adenocarcinoma, SUSD5 was identified as part of a lactylation-related prognostic signature and associated with tumor microenvironment modulation and immune cell infiltration 3. SUSD5 is involved in lung cancer bone metastasis through a dual mechanism: it mediates bone matrix degradation by promoting TGF-beta-dependent osteoclastogenic bone resorption and enhancing metalloproteolytic activities 4. Additionally, SUSD5 was identified in ceRNA regulatory networks in chr3 atrophic gastritis, a precancerous lesion 5, and was elevated in serum of frontotemporal dementia patients, suggesting broader involvement in neuroinflammatory pathways 6. These findings indicate SUSD5's multifaceted roles in cancer progression, metastasis, and immune dysregulation.
