UBE2Q2 is a ubiquitin-conjugating enzyme (E2) located on chromosome 15 that catalyzes the covalent attachment of ubiquitin to target proteins, specifically mediating K48-linked polyubiquitination in vitro 1. The enzyme functions in both nuclear and cytosolic compartments to facilitate protein degradation through the ubiquitin-proteasome system 1. UBE2Q2 exhibits complex and context-dependent roles in cancer. In head and neck squamous cell carcinoma, high UBE2Q2 expression suppresses cell proliferation and is downregulated in chemotherapy-resistant tumors, suggesting an oncosuppressive function 2. Conversely, UBE2Q2 is significantly overexpressed in breast cancer (71.4% of cases), colorectal cancer (65.11% of cases), and acute lymphoblastic leukemia (55% of cases), correlating with poor prognosis 1, 3, 4. In hepatocellular carcinoma, UBE2Q2 promotes tumor progression and glycolysis through NF-κB/HIF1α signaling, where it mediates K63-linked ubiquitination of RIP1 via interaction with cIAP1 5.
