ANKRD46 (ankyrin repeat domain 46) is a protein-coding gene located on chromosome 8-qter that functions as a regulatory target in multiple disease contexts. Mechanistically, ANKRD46 is regulated by microRNAs (miRNAs) including miR-21, miR-25-3p, and miR-451 1 2 3. In the circ_PRDM5/miR-25-3p/ANKRD46 axis, ANKRD46 upregulation suppresses breast cancer cell proliferation, migration, and invasion while enhancing apoptosis 2. During embryo implantation, miR-451-mediated ANKRD46 regulation is critical for successful implantation, as miR-451 inhibition reduces implantation rates 3. ANKRD46 is implicated in cancer drug resistance: it is upregulated in paclitaxel-resistant nasopharyngeal carcinoma cells and is concordantly deregulated in gastric cancer through chr8 amplification 4 5. In lens biology, ANKRD46 is a priority target of dysregulated miRNAs in congenital cataract models, suggesting relevance to lens pathology 6. Clinically, ANKRD46 shows dose-dependent expression changes in post-Chernobyl thyroid cancers associated with radiation exposure 7, and rare loss-of-function variants segregate with alcohol use disorder in multiplex families 8. These findings indicate ANKRD46 functions as a regulatory hub in cancer progression, reproductive biology, and disease susceptibility.
