BHLHA15 is a basic helix-loop-helix transcription factor that functions as a master regulator of secretory cell identity and differentiation across multiple tissues. As a DNA-binding transcription factor, BHLHA15 binds E-box consensus sequences to control gene expression programs 1. In the intestinal epithelium, BHLHA15 marks short-lived secretory precursor cells and Paneth cells; however, these cells can undergo reprogramming into tumor-initiating cells when Notch and YAP signaling are aberrantly activated 2. BHLHA15 plays critical roles in regulating the unfolded protein response (UPR) and exocrine cell function 13. In acinar tissues, BHLHA15 activity is significantly reduced during acinar-to-ductal metaplasia, an early pancreatic cancer development event 4. BHLHA15 expression is upregulated in response to hepatic injury and correlates with liver disease progression through regulation of apoptosis 5. Additionally, BHLHA15 expression increases in aged male skeletal muscle, suggesting involvement in age-related muscle pathology 6. Recent evidence indicates BHLHA15 can be targeted by microRNAs to suppress cancer progression via PTEN/AKT pathway regulation 7.
