CLEC12B encodes a C-type lectin receptor that functions as an inhibitory receptor regulating both immune and non-immune cellular processes. The protein contains an immunoreceptor tyrosine-based inhibitory motif (ITIM) in its intracellular domain that, upon phosphorylation, recruits SH2 domain-containing phosphatases SHP-1 and SHP-2 (PTPN6 and PTPN11) to antagonize activation signals 1 2. CLEC12B is predominantly expressed on skin melanocytes and mast cells, with selective expression patterns across different tissues 3 4. In melanocytes, CLEC12B negatively regulates melanogenesis by promoting CREB degradation and downregulating the MITF pathway, ultimately controlling melanin production and skin pigmentation 3. Additionally, it inhibits melanoma cell proliferation by inactivating STAT3 signaling and increasing p53/p21/p27 expression 2. CLEC12B also plays a role in skin immunity by activating the STAT1/IRF1-IFNγ pathway in response to bacterial recognition, leading to chemokine production and immune cell recruitment 5.
