DCNP1 (dendritic cell nuclear protein 1) is a 244-amino acid nuclear protein specifically expressed in mature dendritic cells with perinuclear localization 1. The protein contains a nuclear localization signal (RRK residues 117-119) required for nuclear translocation 2. DCNP1 functions as a transcriptional regulator: it binds to and transactivates the corticotropin-releasing hormone (CRH) promoter in the hypothalamic paraventricular nucleus 3, and represses N-acetyltransferase (NAT) expression through interaction with the circadian regulator BMAL1 2. These mechanisms suggest DCNP1's involvement in stress response and circadian-regulated melatonin biosynthesis. Clinically, DCNP1 is implicated in major depressive disorder (MDD) pathogenesis, with increased expression in depressed patients' paraventricular nuclei 3; however, genetic association with MDD varies across populations, showing no significant association in Han Chinese cohorts 4. Additionally, DCNP1 promoter polymorphisms associate with allergen-specific IgE responses in asthma, suggesting roles in immune regulation 5. The full-length protein variant appears functionally distinct from truncated forms lacking the nuclear localization signal, with implications for depression and immune-related phenotypes.
