ELP5 (elongator acetyltransferase complex subunit 5) is an essential component of the elongator complex that plays crucial roles in tRNA modification and cellular processes. As part of the Elp4/5/6 subcomplex, ELP5 is required for the integrity and stability of the elongator complex, directly connecting ELP3 to ELP4 1. The elongator complex catalyzes formation of carboxymethyluridine and other modifications in the wobble base at position 34 in tRNAs, which are essential for proper translation 23. ELP5 forms a RecA-ATPase-like fold and contributes to the hexameric ring-shaped structure of the Elp4-6 subcomplex that binds histone H3 4. Functionally, ELP5 regulates cell migration and tumorigenicity in melanoma cells 1. In gallbladder cancer, ELP5 expression is epigenetically silenced through promoter hypermethylation by DNMT3A, blocking PAX5-mediated transcription activation 5. Loss of ELP5 impairs wobble U34 tRNA modification, affecting translation of hnRNPQ mRNA, which subsequently reduces P53-dependent gemcitabine sensitivity in gallbladder cancer 3. ELP5 has also been identified as a pleiotropic gene associated with both schizophrenia and inflammatory bowel disease 6 and as a hub gene in pancreatic ductal adenocarcinoma networks 7.
