GAREM1 (GRB2 associated regulator of MAPK1 subtype 1) functions as an adapter protein that promotes activation of the MAPK/ERK signaling pathway downstream of growth factor receptors, particularly the epidermal growth factor receptor 1. The protein mediates EGF-stimulated cell signaling through tyrosine phosphorylation and Grb2 binding, regulating phosphorylation of downstream effectors including S6 kinase and 4EBP1 necessary for cell proliferation 1. Unlike its brain-specific homolog GAREM2, GAREM1 localizes to the nucleus via a nuclear localization sequence in its N-terminal region 2. GAREM1 plays critical roles in normal growth and body size maintenance; loss-of-function mutations or dominant-negative variants impair ERK activation and are associated with idiopathic short stature in humans and reduced body weight in knockout mice 1.
