IRAG2 (also known as Jaw1/LRMP) is a membrane protein that functions as a regulator of intracellular calcium signaling and cellular structure. As a cGMP kinase substrate protein, IRAG2 modulates calcium release by enhancing Ca2+ release through interaction with inositol 1,4,5-trisphosphate receptors (ITPRs), particularly accelerating signal onset time and rise time in cells expressing ITPR1 1. The protein localizes to the endoplasmic reticulum and outer nuclear membrane, where it functions as a KASH protein to maintain nuclear shape through microtubule interactions 2. IRAG2 also plays a crucial role in maintaining Golgi ribbon structure, as its depletion causes Golgi fragmentation with disordered ribbon structure 2.
