LRRC19 is a leucine-rich repeat-containing transmembrane pathogen-recognition receptor that activates innate immunity through TRAF2- and TRAF6-mediated NF-κB signaling 1. In kidney, LRRC19 prevents uropathogenic Escherichia coli infection by inducing production of cytokines, chemokines, and antimicrobial substances 1. In the gut, LRRC19 plays a complex dual role: it responds to pathogenic signals, notably interacting with Desulfovibrio vulgaris flagellin (DVF) to promote pro-inflammatory cytokine production and exacerbate colitis 2, while simultaneously degrading tight junction proteins (ZO-1, ZO-3, occludin) via PKCζ and PKCι/λ ubiquitination to increase gut barrier permeability 3. In colorectal cancer, LRRC19 expression is significantly downregulated and associated with poor prognosis 4. However, when restored, LRRC19 suppresses CRC progression by inhibiting CDK6/E2F1-mediated cell cycle progression, enhancing immune infiltration of protective B and CD4+ T cells, and sensitizing cells to oxaliplatin chemotherapy 5. These findings establish LRRC19 as a context-dependent immunoregulator with therapeutic potential targeting the DVF-LRRC19 axis in inflammatory bowel disease and the CDK6/E2F1 pathway in colorectal cancer.
