While originally characterized in B cells, recent evidence reveals that NFAM1 is highly expressed in monocytes and neutrophils rather than lymphocytes 2. In monocytes, NFAM1 amplifies pro-inflammatory cytokine production (TNF-α, IL-6, IL-12) and chemokine responses (CCL3, CCL4) to diverse stimuli including CD40L and TLR ligands 2. NFAM1 regulates monocyte chemotaxis through p38 MAPK signaling and chemokine receptor expression 3. Disease relevance includes inflammatory bowel disease, where NFAM1 expression is significantly elevated in affected tissues 2, allergic diseases including atopic dermatitis 4, coronary artery disease, where increased monocytic NFAM1 correlates with atherosclerotic burden 3, acute myocardial infarction through dendritic cell maturation 5, asthma exacerbation 6, and Paget's disease of bone where viral-induced NFAM1 drives osteoclast differentiation 7.
