RAVER2 (ribonucleoprotein, PTB binding 2) is an RNA-binding protein and member of the heterogeneous nuclear ribonucleoprotein (hnRNP) family that functions as a modulator of polypyrimidine-tract binding protein (PTB) activity 1. The protein contains three RNA recognition motifs and displays nucleo-cytoplasmic shuttling, localizing primarily to the nucleus in neural tissues 2. RAVER2 interacts directly with PTB and regulates alternative splicing of PTB-target genes, including the myelin protein PLP1 3. While human RAVER2 is ubiquitously expressed across adult tissues, its expression is tightly regulated through TATA-less transcriptional mechanisms and is dependent on lamin B1 levels in neural tissue 13. Clinically, RAVER2 dysregulation is implicated in multiple disease pathways. RAVER2 overexpression contributes to autosomal dominant leukodystrophy (ADLD) by altering splicing patterns of neurologically important genes 3. Genetic variants in RAVER2 are associated with increased susceptibility to ulcerative colitis 4 and methotrexate-induced hepatotoxicity in rheumatoid arthritis patients 5. Additionally, RAVER2 appears dysregulated in colorectal cancer as a microRNA target 6 and may function in viral immune responses during Zika virus infection 7.
