RIPOR2 (RHO family interacting cell polarization regulator 2) is a multifunctional protein that regulates cell polarity and immune responses. Functionally, RIPOR2 acts as an inhibitor of the small GTPase RHOA 1 and promotes filopodia and myotube formation in myoblast differentiation 2. The protein maintains naive T lymphocytes in a quiescent state 3 and has emerged as a critical immunometabolic regulator that prevents T cell exhaustion; RIPOR2 depletion exacerbates exhaustion phenotypes with reduced cytokine production 4. Mechanistically, RIPOR2 localizes to stereocilia of auditory hair cells where it interacts with Myh9 and influences protein abundance and post-translational modifications in both stereocilia and kinocilia 5. The protein regulates cell polarization through negative regulation of Rho signaling and affects immune cell migration and chemotaxis 4. Clinically, RIPOR2 mutations cause non-syndromic cochleovestibular dysfunction with congenital bilateral profound hearing loss and vestibular dysfunction 6. RIPOR2 acts as a protective tumor-suppressor biomarker; decreased expression is associated with poor prognosis in cervical cancer, correlating with genomic instability and unfavorable immunotherapy response 78.
