The protein contains a conserved GTPase domain and a carboxy-terminal coiled-coil domain characteristic of septins, and forms heterodimeric complexes with other septins like SEPT7 and SEPT9 through parallel coiled-coil interactions 21. SEPTIN14 is predominantly expressed in testis tissue, where it plays a critical role in spermatogenesis and sperm head formation during spermiogenesis 34. Specifically, SEPTIN14 protein localizes to the sperm acrosome and neck region, and reduced expression correlates with spermatogenic failure, hypospermatogenesis, and maturation arrest 4.
