Based on limited published evidence, TMCO6 is a transmembrane protein with coiled-coil domains that functions as a protein-binding receptor. The N-terminus of TMCO6 binds neutrophil extracellular trap (NET) DNA on CD8+ T cells, suppressing T-cell receptor signaling and NFκB p65 nuclear translocation to impair antitumor immunity in hepatocellular carcinoma 1. TMCO6 methylation patterns are associated with periodontal disease phenotypes in adult twins 2, suggesting broader immunological roles.
