Based on limited published evidence, WASHC2A is a core component of the WASH complex that localizes to endosomes and regulates actin polymerization and vesicle trafficking. It binds phospholipids (particularly PtdIns-(4)P, PtdIns-(5)P, and PtdIns-(3,5)P2) and recruits the WASH complex to endosomal membranes via interaction with retromer VPS35, inhibiting Arp2/3-mediated actin nucleation to promote tubule fission during endosomal sorting. WASHC2A mediates endosome-to-plasma membrane trafficking of SNX27-retromer cargo (e.g., GLUT1) and recruits F-actin-capping proteins and CCC/retriever complex components. A circular RNA isoform of WASHC2A shows potential as a prognostic biomarker in pediatric B-ALL 1.
