ZGLP1 is a zinc finger transcription factor with dual roles in germ cell development and vascular pathology. In germ cells, ZGLP1 functions as a master determinant of oogenic fate by activating genes required for the oogenic program and meiotic prophase entry 1. It operates downstream of bone morphogenetic protein signaling but independently of retinoic acid, regulating expression of STRA8 and other oogenic effectors while antagonizing Polycomb-mediated repression 1. ZGLP1 overexpression drives differentiation of primordial germ cell-like cells into fetal oocytes in vitro 1. In males, ZGLP1 is required for spermatogonia to efficiently complete meiotic prophase 1. Recently, ZGLP1 was identified as one of five transcription factors sufficient to rapidly convert human induced pluripotent stem cells to oogonia-like cells within four days in feeder-free culture 2. Beyond reproduction, ZGLP1 regulates vascular smooth muscle cell homeostasis; ZGLP1 knockdown promotes apoptosis and phenotypic transformation from contractile to synthetic phenotype via the protein kinase B pathway 3, suggesting potential involvement in intracranial aneurysm pathogenesis. These findings establish ZGLP1 as both a key regulator of human in vitro gametogenesis and a potential therapeutic target for vascular diseases.
