CCDC178 (coiled-coil domain containing 178) is an oncogenic protein with emerging roles in cancer progression and metastasis. While UniProt annotations suggest involvement in spermatogenesis and centrosomal function, recent cancer biology research reveals CCDC178's primary pathological function involves promoting metastatic traits through ERK pathway activation. In hepatocellular carcinoma, CCDC178 upregulation correlates with pathological stage and promotes anoikis resistance and metastatic potential by associating with BRAP2, a negative regulator of ERK signaling, and promoting its degradation 1. Similarly, in gastric cancer, high CCDC178 expression predicts poor prognosis and recurrent disease 2. CCDC178 can be transcriptionally activated by BARX1, and this axis activates ERK pathway signaling to drive gastric cancer cell migration, invasion, and tumor-associated macrophage M2-like polarization 3. CCDC178 also appears in fusion oncoproteins: a PIK3R1-CCDC178 fusion identified in ovarian cancer promotes cell migration and platinum chemoresistance through ERK1/2 activation and formation of rod and ring-like cellular structures 4. Additionally, CCDC178 mutations have been identified in gastric cancer with peritoneal metastasis 5, and elevated CCDC178 expression appears as a biomarker in inflammatory pulp pathology 6. These findings position CCDC178 as a novel therapeutic target in metastatic cancers, though its normal physiological functions remain incompletely characterized.
