DIMT1 (DIM1 rRNA methyltransferase and ribosome maturation factor) is an RNA methyltransferase that catalyzes N6,6-dimethyladenosine (m6,6A) modifications in 18S rRNA within the 40S ribosomal subunit 1. Beyond its catalytic function, DIMT1 participates in pre-rRNA processing and ribosome biogenesis as a component of the small subunit (SSU) processome 23. Notably, DIMT1 exerts dual functions: its methyltransferase activity modifies both 18S rRNA and small RNAs 4, while its noncatalytic scaffold function is essential for rRNA binding, nucleolar localization via liquid-liquid phase separation, and ribosome assembly independent of catalytic activity 5. Clinically, DIMT1 dysfunction impairs protein synthesis and ribosome maturation. DIMT1 deficiency disrupts β-cell mitochondrial function and insulin secretion, implicating it in type 2 diabetes pathogenesis 6. Conversely, DIMT1 overexpression correlates with malignancy: elevated expression predicts poor prognosis in gastric carcinoma 7 and is essential for acute myeloid leukemia proliferation 5. DIMT1 is upregulated in multiple myeloma, where microRNA-210 suppresses it under hypoxic conditions 8. Additionally, DIMT1 overexpression extends lifespan and protects cells from senescence 9, suggesting potential longevity regulation through oxidative phosphorylation pathways.
