LRP11 (LDL receptor-related protein 11) is a phosphoprotein-binding protein involved in lipid metabolism and cancer progression. Functionally, LRP11 facilitates lipid accumulation in hepatocellular carcinoma by recruiting USP5 to enhance deubiquitination and stabilization of RACK1 1, with critical interaction domains localized to residues 309-500. Beyond lipid metabolism, LRP11 promotes proliferation in lung adenocarcinoma through transcriptional modulation of C/EBPβ-binding genes and shows high expression correlating with poor clinical prognosis 2. In prostate cancer, LRP11 activates β-catenin signaling to induce PD-L1 expression, facilitating immune evasion 3. The long non-coding RNA LRP11-AS1 mediates enterotoxigenic Bacteroides fragilis-induced colorectal carcinogenesis via miR-149-3p/CDK4 pathway regulation 4. Clinically, LRP11 shows broad disease relevance: genetically determined higher LRP11 levels associate with increased hippocampal volume and reduced Alzheimer's disease risk 5, while LRP11 represents a potentially causal protein in chr6 kidney disease progression with druggable potential 6. Environmental exposure to per- and polyfluoroalkyl substances associates with altered LRP11 protein levels in cardiometabolic pathways 7. These findings establish LRP11 as a multifunctional protein with therapeutic potential across multiple malignancies and neurodegenerative diseases.
