NANOGP8 is a protein-coding retrogene that functions as a transcription factor with cell-type-specific expression patterns 1. Although originally classified as a pseudogene, NANOGP8 encodes a functional protein nearly identical to embryonic NANOG (differing by only three amino acids) and exhibits comparable reprogramming activity in converting fibroblasts to induced pluripotent stem cells 2. In normal cells, NANOGP8 expression is generally restricted to specific cell types including smooth muscle cells 1, but it is widely expressed across multiple cancer types. Oncologically, NANOGP8 promotes malignant phenotypes through multiple mechanisms. In gastric cancer, NANOGP8 is overexpressed in 75% of tumors compared to 12.5% of adjacent normal tissues 3, and activates the AKT/mTOR signaling pathway to enhance cell proliferation and migration 4. In prostate cancer, NANOGP8 knockout via CRISPR/Cas9 significantly reduces sphere formation, anchorage-independent growth, migration, and drug resistance, though not in vitro proliferation 5. NANOGP8 expression is transcriptionally regulated by the ETS family factor Fli-1 in leukemia 6. Beyond nuclear transcriptional roles, NANOGP8 localizes at centrosomes and associates with centriole maturation across tumor and non-tumor cells 7.
