OR4C12 is an olfactory receptor family member located on chromosome 11 that functions as a G-protein coupled receptor with roles extending beyond classical olfactory signaling. In pancreatic cells, OR4C12 acts as a receptor for R-(-)-carvone, activating GNAS-mediated signaling pathways that increase intracellular Ca²⁺ and cAMP levels, ultimately promoting glucose-stimulated insulin secretion. This suggests a potential role in glucose homeostasis regulation. Genetically, OR4C12 variants show associations with multiple physiological processes. A genome-wide association study identified OR4C12 as significantly associated with circulating CD34⁺ progenitor cell frequency in a large community-based cohort 1, suggesting involvement in hematopoietic stem cell regulation. OR4C12 has been implicated in chr11 dry cough and ACEi-induced cough susceptibility through genetic association studies mapping neurological pathways 2, indicating potential involvement in sensory neuronal function. Additionally, OR4C12 loss (chromosome 11.12) was identified in approximately 48% of primary CNS PNETs and 50% of primary pineoblastomas 3, suggesting possible tumor suppressor properties in embryonal brain tumors.
