PPP1R2C is a protein phosphatase inhibitor that functions as a regulator of type-1 protein phosphatase (PP1) activity. The protein inhibits the catalytic subunit of PP1 and weakly inhibits myosin-associated phosphatases through direct binding interactions. PP1 regulators like PPP1R2C emerged early in eukaryotic evolution as primordial determinants of PP1 function, with vertebrate lineages experiencing a marked expansion of PP1 regulatory proteins 1. In cardiac tissue, inhibitor-1 (a related family member) functions as a nodal integrator of cAMP and calcium signaling pathways, with phosphorylation at multiple sites modulating PP1 activity and contractility 2. PPP1R2C is associated with diverse disease contexts including amyotrophic lateral sclerosis, colorectal carcinoma, germ cell tumors, Graves disease, lung carcinoma, and sepsis, though mechanistic links remain largely unexplored. At the clinical level, polymorphisms in related inhibitor-1 sequences show variable associations with heart failure outcomes depending on genetic background, with the G147D variant identified primarily in Black populations acting as a potential disease modifier rather than primary risk factor 3. Further work is needed to establish PPP1R2C-specific roles in these pathological contexts.
