SCART1 is a scavenger receptor cysteine-rich (SRCR) superfamily member functioning as a co-receptor on T lymphocytes. Structurally, human SCART1 contains five extracellular SRCR domains, a transmembrane region, and a cytoplasmic domain with putative phosphorylation sites and internalization motifs 1. In mice, SCART1 is expressed on approximately 86% of γδ T cells but absent from αβ CD4+ and CD8+ T cells 2. Human SCART1 shows broader expression on both CD4+ and CD8+ T lymphocytes with either αβ or γδ T cell receptors 1, with high tissue expression in small intestine and colon 1. Functionally, SCART1 likely serves as a pattern recognition receptor and activating co-receptor on γδ T cells, though it does not bind selected bacterial or fungal components directly 2. SCART1+ cell numbers are significantly elevated in inflammatory conditions: an 8.6-fold increase in skin inflammation and elevated numbers in inflammatory bowel disease 2, suggesting a role in pathogenic immune responses. SCART1's structural relationship to the WC1 family indicates participation in γδ T cell-mediated immunity 3, though specific molecular interactions remain incompletely characterized.
