ANAPC13 is a component of the anaphase promoting complex/cyclosome (APC/C), a cell cycle-regulated E3 ubiquitin ligase that mediates progression through mitosis and G1 phase. The APC/C catalyzes ubiquitination and degradation of cell cycle regulatory proteins, primarily through assembly of Lys-11-linked polyubiquitin chains, with secondary contributions from Lys-48- and Lys-63-linked chains. ANAPC13 dysfunction has been implicated in multiple disease contexts. In a mouse model with regulatory T cell-specific SF3B1-K700E mutation, aberrant splicing reduced ANAPC13 expression, resulting in defective Treg differentiation and impaired colitis prevention; forced ANAPC13 expression restored these immune functions 1. ANAPC13 has been identified as a differentially expressed gene in oral cancer progression networks 2 and in studies of okadaic acid-induced cellular toxicity in neuroblastoma cells 3. A non-coding somatic mutation disrupting HNF4α binding in liver cancer was found to affect an enhancer regulating ANAPC13 alongside other liver cancer-associated genes 4. Additionally, ANAPC13 DNA methylation in nasal epithelium was associated with atopic asthma in a meta-analysis of three pediatric cohorts 5. These findings suggest ANAPC13 dysregulation contributes to both immune dysfunction and cancer progression across multiple tissue contexts.
