PALM3 (paralemmin 3) is an ATP-binding adapter protein that plays a critical role in Toll-like receptor (TLR) signaling and inflammatory responses. Functionally, PALM3 acts as an adaptor in LPS-TLR4 signaling pathways 1, interacting with key signaling molecules including TLR4, myeloid differentiation factor 88, IL-1 receptor-associated kinase-1, TNF receptor-associated factor-6, and Toll-IL-1 receptor-containing adapter molecule-2 2. LPS stimulation upregulates PALM3 expression in alveolar epithelial cells and macrophages 12, and PALM3 knockdown suppresses LPS-induced cytokine release and inflammatory responses 12, suggesting PALM3 promotes rather than inhibits inflammatory signaling. Clinically, PALM3 has emerged as relevant to multiple disease contexts. It was identified as a novel age-related hearing loss gene in genome-wide association studies 3, and shows prognostic significance in colon cancer, where PALM3 expression associates with patient outcomes 4. Additionally, PALM3 was incorporated into a glycolysis-metabolism-related prognostic signature for Ewing sarcoma 5. These findings suggest PALM3 participates in both immune inflammatory pathways and disease progression mechanisms across diverse conditions.
