PCDHB2 (protocadherin beta 2) is a calcium-dependent cell adhesion molecule involved in establishing and maintaining neuronal connections. The protein localizes to the plasma membrane and functions in cell-cell adhesion and synapse assembly during nervous system development 1. As a member of the cadherin superfamily, PCDHB2 mediates calcium-dependent cell adhesion through cell adhesion molecule binding interactions. Its expression is particularly relevant in neural tissue, where it supports the establishment of specific neuronal connections critical for proper brain development and function. Notably, PCDHB2 expression shows sex-specific regulation in the adult cerebral cortex, with elevated expression observed in female offspring following fetal alcohol exposure compared to males 1. Beyond its developmental role, PCDHB2 has emerged as a clinically relevant biomarker in multiple cancer contexts. It was identified as one of nine m6A-related genes associated with colorectal cancer prognosis and overall survival 2, and as a differentially expressed gene in radiotherapy response prediction for HPV-negative cervical cancer 3. PCDHB2 was also included in a prognostic signature for cyclophosphamide response in breast cancer 4, and identified as differentially expressed in cribriform pattern prostate cancer 5. Additionally, PCDHB2 methylation patterns associate with arsenic-induced urothelial carcinoma 6, and potential driver mutations in pancreatic cancer 7. These cancer associations suggest PCDHB2 alterations may contribute to tumor progression and treatment response.
