PNLIPRP2 encodes a pancreatic lipase that catalyzes hydrolysis of multiple lipid substrates. It cleaves triglycerides containing short-, medium-, and long-chain fatty acyls, and can completely deacylate triacylglycerols [UniProt]. In neonates, PNLIPRP2 plays a major role in digesting dietary fats from milk, particularly long-chain triglycerides [UniProt]. As the liver matures, the enzyme's function shifts to primarily hydrolyzing galactolipids—plant-derived molecules including monogalactosyldiglycerol (MGDG) and digalactosyldiacylglycerol (DGDG)—releasing long-chain polyunsaturated fatty acids [UniProt]. PNLIPRP2 also exhibits monoacylglycerol lipase and low phospholipase activities [UniProt]. In pancreatic ductal adenocarcinoma (PDAC), PNLIPRP2 expression is significantly reduced compared with adjacent nontumor tissues, consistent with impairment of lipolytic pathways 1. During acute pancreatitis, PNLIPRP2 and pancreatic triacylglycerol lipase are equipotent in inducing lipolysis and lipotoxic injury in fat necrosis 2. A common truncation variant (p.W358X) is expressed poorly and does not increase risk for chr10 pancreatitis 3. Genome-wide association studies link PNLIPRP2 variants to phytosterol and phospholipid metabolism, with sitosterol concentrations having a causal relationship with coronary atherosclerosis risk 45.
