RTP1 (receptor transporter protein 1) is a molecular chaperone that specifically promotes functional cell surface expression of olfactory receptors 1. As a receptor transporter protein, RTP1 facilitates protein insertion into cellular membranes and enhances receptor trafficking to the cell surface 2. In vitro studies demonstrate that RTP1 increases surface expression of chemosensory channels, such as TRPC2, and correspondingly enhances cellular signaling capacity in patch-clamp electrophysiological assays 2. Beyond its primary olfactory function, RTP1 is broadly expressed throughout the central nervous system and shows altered expression patterns in multiple neurodegenerative diseases including Parkinson's disease, Alzheimer's disease, and Creutzfeldt-Jakob disease 1. RTP1 has emerged as a disease-relevant gene in neurological conditions; whole-exome sequencing identified de novo RTP1 mutations in West syndrome patients with epileptic encephalopathy, and network analyses revealed tight genetic associations with known epilepsy genes 3. Additionally, genetic variants in RTP1 show suggestive association with xerostomia susceptibility in oropharyngeal cancer survivors 4, and RTP1 has been identified as a potential biomarker regulating osteosarcoma prognosis 5. These findings indicate RTP1's broader physiological importance beyond olfactory receptor trafficking.
