RTP3 (receptor transporter protein 3) is a membrane-targeting protein that promotes functional cell surface expression of bitter taste receptors, particularly TAS2R16 and TAS2R43 1. The protein facilitates chemoreceptor trafficking through direct protein-protein interaction with taste receptors, enabling their localization from the Golgi compartment to the cell membrane 1. RTP3 is expressed in chemosensory tissues including circumvallate papillae and testis 1. Beyond taste sensation, RTP3 has emerged as a candidate gene for bone homeostasis. A genome-wide association study identified SNPs in RTP3 associated with femoral neck bone geometry parameters, including buckling ratio and cortical thickness, and with hip fracture risk 2. Recent evidence suggests RTP3 participates in cancer-related signaling: RTP3 expression modulates the STAT3 pathway in hepatocellular carcinoma progression 3, and RTP3 was identified as one of 17 key genes in a diagnostic model for metabolic dysfunction-associated steatotic liver disease 4. Additionally, RTP3 expression is significantly downregulated in clear cell renal cell carcinoma and may serve as a clinical descriptor of advanced tumors 5. These findings suggest RTP3 functions extend beyond chemoreceptor trafficking to include roles in bone metabolism and tumor suppression.
