SCAPER (S-phase cyclin A-associated protein in the ER) is a cell cycle regulatory protein that functions as a cyclin A/Cdk2 binding partner, transiently sequestering this kinase in the cytoplasm and endoplasmic reticulum to regulate cell cycle progression 1. Beyond its classical cell cycle role, SCAPER localizes to primary cilia where it regulates microtubule dynamics and ciliary disassembly 2. SCAPER also recruits the CCR4-NOT deadenylase complex to ribosomes through interaction with CNOT11, coupling nascent tubulin polypeptide recognition to mRNA decay and enabling proper microtubule-dependent chromosome 15 3. SCAPER is expressed during neural development with higher levels in neurons and shows layer-specific expression in the adult cerebral cortex 4. Biallelic SCAPER mutations cause autosomal recessive intellectual developmental disorder with retinitis pigmentosa (IDDRP), characterized by visual degeneration, cognitive impairment, and additional features including dysmorphia, ciliary dysfunction, and skeletal anomalies 56.
