SNRNP25 (small nuclear ribonucleoprotein U11/U12 subunit 25) is a structural component of the minor spliceosome that catalyzes U12-dependent intron splicing. Structurally, SNRNP25 specifically recognizes and stabilizes U11 small nuclear RNA (snRNA) within the 13-subunit U11 snRNP particle complex 1. It functions as part of a network where PDCD7 bridges SNRNP25 to SNRNP48, positioning these factors to facilitate 5' splice site recognition through base-pairing interactions with U11 snRNA 1. Clinically, SNRNP25 dysfunction has multiple disease implications. A recurrent LRP1-SNRNP25 fusion gene is osteosarcoma-specific and promotes tumor cell migration and invasion in osteosarcoma models 23. Additionally, SNRNP25 loss is implicated in early mammalian development, as disruption of the Snrnp25 locus in mice correlates with peri-implantation lethality in homozygous deletions 4. SNRNP25 has also emerged as an autoantigen in scleroderma patients without standard autoantibody specificities, suggesting its role in disease-associated immune responses 5. These findings highlight SNRNP25's dual significance in RNA splicing machinery and disease pathogenesis.
